Hemileia vastatrix, agente causal de la roya del café, es un parásito obligado. La preservación en laboratorio bajo condiciones de frío (rangos de 2-8 °C) puede reducir la germinación y viabilidad de las uredosporas por tiempos prolongados. Esto es una limitante al momento de realizar investigaciones in vitro. Objetivo. Determinar el tiempo viable de conservación de uredosporas en frío, mediante la evaluación del porcentaje de germinación, utilizando dos dispersantes para la germinación de forma artificial en medio de cultivo. Materiales y métodos. Se evaluó cuatro períodos de 22 días de diferencia entre cada uno de los tratamientos, se preservó inóculo de roya a temperatura de 4 y -30 °C, por ser temperaturas utilizadas en el almacenaje de fitopatógenos en diferentes laboratorios. La dispersión de uredosporas se realizó de manera in vitro en placas Petri con medio de cultivo agar-agua, se utilizó agua desionizada y Tween® 20 como dispersante. Se incubaron las placas por 12 horas a 21 °C y se realizó el conteo de uredosporas viables. Resultados. Se obtuvo un porcentaje de germinación a los 0 días después de almacenamiento (DDA) de 58,0% con Tween® 20 y 36,2% con agua desionizada. Realizadas las cuatro evaluaciones, se determinó que la temperatura de almacenamiento a 4 °C mantiene constante la germinación hasta los 88 DDA, mientras que el inóculo preservado a -30 °C redujo significativamente la germinación de uredosporas a partir de los 44 DDA. El uso del dispersante Tween® 20 mantuvo un porcentaje de germinación superior en los períodos de 22, 44 y 88 DDA, al tratamiento de agua desionizada.Conclusión. El Tween® 20 genera una mayor dispersión de la uredosporas, lo que inhibe la germinación. El choque térmico de los uredosporas, al pasar de temperaturas ambientales cálidas a preservación a -30 °C, disminuye la germinación in vitro.]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[Abstract Introduction. Hemileia vastatrix, the causal agent of coffee rust, is an obligatory parasite. Preservation in the laboratory under cold conditions (ranges of 2-8 °C) can reduce the germination and viability of uredospores for extended periods, which is a limiting factor when conducting in vitro research. Objective. To determine the viable storage time of uredospores under cold conditions by evaluating the germination percentage, using two dispersants for artificial germination in a culture medium. Materials and methods. Four periods, 22 days apart, were evaluated for each treatment. Rust inoculum was preserved at temperatures of 4 °C and -30 °C, as these are commonly used temperatures for storing phytopathogens in different laboratories. Uredospores were dispersed in vitro on Petri dishes with agar-water culture medium, using deionized water and Tween 20 as dispersants. The plates were incubated for 12 hours at 21 °C, and the number of viable uredospores was counted. Results. A germination percentage of 58.0% with Tween 20 and 36.2% with deionized water was obtained on the 0 days after storage (DAS). After the four evaluations, it was determined that the storage temperature at 4 °C maintained constant germination up to 88 DAS, while the inoculum preserved at -30 °C significantly reduced uredospore germination from 44 DAS onward. The use of Tween 20 dispersant maintained a higher germination percentage at the 22, 44 and 88 DAS periods compared to the deionized water treatment. Conclusion. Tween 20 generates greater dispersion of the uredospores, which inhibits germination. The thermal shock to the uredospores when transitioning from warm ambient temperatures to preservation at -30 °C reduces in vitro germination.]]></p></abstract>
