No se encontraron diferencias estadísticamente significativas entre las frecuencias esperadas y las observadas, de acuerdo a la ley de Hardy Weimberg. Los resultados de Glo I, EsD y EAP son comparables a los encontrados en poblaciones caucásicas norteamericanas y AK con poblaciones hispánicas norteamericanas. Por medio del cálculo del poder de discriminación, se valoró su utilidad en el campo forense como marcadores de individualidad. El poder de discriminación con valores superiores a .34 encontrado en la EsD, Glo I, PGM sub, EAP y Gc indica que éstos marcadores son una herramienta probabilística útil para la Administración de Justicia, en donde la sangre u otro fluido corporal son un indicio.]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[555 blood samples from different areas of the Costa Rican territory was studied by electrophoretic analysis to determine the phenotypic and genotypic frequencies of 10 polymorphic allozimes (PGM sub, EsD, AK, EAP, Glol, ADA, CA II, PepA, Gc y Tf). There are no significant differences between the expected distribution and the ones found according to the Hardy-Weimberg Law. The results are comparable with other population studies that include North American Caucasian and Hispanic populations. The calculation of the discrimination power is useful in forensic science and the values obtained in EsD, Glo I, PGM sub EAP and Gc make these markers an useful probabilistic tool in the Administration of Justice.]]></p></abstract>

The calculation of the discrimination power is useful in forensic science and the values obtained in EsD, Glo I, PGM sub EAP and Gc make these markers an useful probabilistic tool in the Administration of Justice.</FONT></FONT>     <BR><FONT SIZE=-1></FONT>&nbsp;<FONT SIZE=-1></FONT>      <P><B><FONT FACE="Arial,Helvetica"><FONT SIZE=-1>Introducci&oacute;n</FONT></FONT></B><FONT SIZE=-1></FONT>      <P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">Las izosimas o isoenzimas son convenientemente definidas como formas moleculares m&uacute;ltiples de una enzima dada que act&uacute;an sobre el mismo sustrato y catalizan la misma reacci&oacute;n, se pueden presentar en un mismo individuo o en diferentes miembros de una misma especie y algunas de ellas se han definido como marcadores gen&eacute;ticos porque los genes para una isoenzima particular est&aacute;n estrechamente relacionados a un locus gen&eacute;tico y la variaci&oacute;n en la poblaci&oacute;n es una indicaci&oacute;n de una simple variaci&oacute;n gen&eacute;tica Mendeliana (<A HREF="#1">1</A>). El mayor objetivo del uso de marcadores gen&eacute;ticos en sangre o fluidos corporales en investigaciones m&eacute;dico-legales, es simplemente establecer la identidad o no identidad entre muestras analizadas y comparadas (<A HREF="#2">2</A>).</FONT></FONT><FONT SIZE=-1></FONT>      <P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">Se define poder de discriminaci&oacute;n como la probabilidad de que dos individuos escogidos al azar presenten formas isoenzim&aacute;ticas diferentes en un sistema dado. Mediante electroforesis, se logra la separaci&oacute;n e identificaci&oacute;n de las formas isoenzim&aacute;ticas presentes en una persona o muestra y de esta forma se establecen las frecuencias genot&iacute;picas y fenot&iacute;picas de cada una de ellas en la poblaci&oacute;n.(<A HREF="#2">2</A>)</FONT></FONT><FONT SIZE=-1></FONT>      <P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">Algunas isoenzimas se han utilizado ampliamente en las investigaciones forenses o m&eacute;dico legales por presentar mucha variabilidad entre las personas, mantener su expresi&oacute;n a lo largo de la vida, ser estables si se almacenan adecuadamente, ser de herencia codominante, su determinaci&oacute;n se realiza</FONT> <FONT FACE="Arial,Helvetica">por procedimientos simples que consumen poca muestra e idealmente el genotipo se puede inferir directamente del fenotipo.(<A HREF="#3">3</A>).</FONT></FONT><FONT SIZE=-1></FONT>      ]]></body>
<body><![CDATA[<P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">Las isoenzimas eritrocitarias m&aacute;s utilizadas en la investigaci&oacute;n forense son: subtipeo de la Fosfoglucomutasa (PGMsub), Adenilato Quinasa (AK), Fosfatasa Acida Eritrocitaria (EAP), Glioxilasa (GLO), Esterasa D (EsD), Adenosindeaminasa (ADA), Anhidrasa carb&oacute;nico (CA), Peptidasa A (PepA). Tambi&eacute;n tenemos los llamados sistemas de grupo s&eacute;ricos donde se incluyen el Componente Grupo Espec&iacute;fico (Gc) y Transferrina (Tf) (<A HREF="#3">3</A>).</FONT></FONT><FONT SIZE=-1></FONT>      <P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">Por tratarse de marcadores gen&eacute;ticos adem&aacute;s de su uso en casos penales, tambi&eacute;n pueden utilizarse en investigaciones de paternidad (<A HREF="#4">4</A>).</FONT></FONT><FONT SIZE=-1></FONT>      <P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">En nuestro pa&iacute;s la determinaci&oacute;n rutinaria de marcadores isoenzim&aacute;ticos en ciencias forenses se dio al inicio de los a&ntilde;os 90's, pero al carecer de estudios poblacionales, &uacute;nicamente se emitieron dict&aacute;menes con criterios excluyentes o no excluyentes carentes de datos estad&iacute;sticos.</FONT></FONT>     <BR><FONT SIZE=-1>&nbsp;</FONT>     <BR><FONT SIZE=-1>&nbsp;</FONT>     <BR><B><FONT FACE="Arial,Helvetica"><FONT SIZE=-1>Materiales y m&eacute;todos</FONT></FONT></B><FONT SIZE=-1></FONT>      <P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">Se analizan 555 muestras de sangre pertenecientes a individuos de la poblaci&oacute;n general de Costa Rica, adultos no emparentados, provenientes de diferentes zonas del pa&iacute;s que acudieron a los laboratorios forenses por diferentes motivos.</FONT></FONT><FONT SIZE=-1></FONT>      <P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">Se prepararon manchas de suero y de sangre en cuadros de tela de algod&oacute;n prelavados, utilizando suero y 1 ml. de</FONT> <FONT FACE="Arial,Helvetica">sangre total. Se rotularon adecuadamente y se secaron a temperatura ambiente para luego mantenerlas a -20&deg;C (Congelador Seco lsotemp Fisher Scientific).</FONT></FONT><FONT SIZE=-1></FONT>      <P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">De las manchas se corta un trozo de tela de aproximadamente 4mm2 y se extrae durante 15 a 30 minutos con reactivo de Cieland (Ditriotreitol -DTT 0.05M); los extractos son utilizados para humedecer hebras de algod&oacute;n prelavado o bien se pueden sacar directamente hebras de las telas con las manchas y humedecerlas en DTT por un lapso de tiempo igual.</FONT></FONT><FONT SIZE=-1></FONT>      <P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">El an&aacute;lisis se realiz&oacute; por medio de m&eacute;todos electrofor&eacute;ticos convencionales en c&aacute;maras horizontales SERI E005 a 5&deg;C (Enfriador RTE 110) con geles de agarosa y/o almid&oacute;n y utilizando peque&ntilde;as cantidades del extracto (<A HREF="#5">5</A>).</FONT></FONT><FONT SIZE=-1></FONT>      ]]></body>
<body><![CDATA[<P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">La mayor&iacute;a de las electroforesis en Ciencias Forenses son de zona e involucran el desplazamiento de las prote&iacute;nas en forma de manchas o bandas zonales (<A HREF="#6">6</A>). Las corridas electrofor&eacute;ticas se realizaron en grupos isoenzim&aacute;ticos que comparten los mismos requerimientos en cuanto a la diferencia de potencial aplicada al medio, la fuerza i&oacute;nica y viscosidad del medio, la temperatura y la masa molecular de la prote&iacute;na (<A HREF="#5">5</A>).</FONT></FONT>     <BR><FONT SIZE=-1></FONT>&nbsp;<B><FONT FACE="Arial,Helvetica"><FONT SIZE=-1></FONT></FONT></B>      <P><FONT SIZE=-1><B><FONT FACE="Arial,Helvetica">Grupo I:</FONT></B></FONT><FONT SIZE=-1></FONT>      <P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">EsD y Giol: Gel agarosa-almid&oacute;n 2%, 400V por 2 horas 20 minutos.</FONT></FONT><FONT SIZE=-1></FONT>      <P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">PGMsub. Gel agarosa 1%, 450V por 4 horas.</FONT></FONT>     <BR><FONT SIZE=-1></FONT>&nbsp;<B><FONT FACE="Arial,Helvetica"><FONT SIZE=-1></FONT></FONT></B>      <P><FONT SIZE=-1><B><FONT FACE="Arial,Helvetica">Grupo lI:</FONT></B></FONT><FONT SIZE=-1></FONT>      <P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">EAP, ADA y AK. <body><![CDATA[<P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">Gc y Tf: gel de agarosa 1%, 400V por 1-2 horas.</FONT></FONT>     <BR><FONT SIZE=-1></FONT>&nbsp;<B><FONT FACE="Arial,Helvetica"><FONT SIZE=-1></FONT></FONT></B>      <P><FONT SIZE=-1><B><FONT FACE="Arial,Helvetica">Grupo IV:</FONT></B></FONT><FONT SIZE=-1></FONT>      <P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">CA II y PepA: Agarosa 1%, 400V por 2 horas.</FONT></FONT><FONT SIZE=-1></FONT>      <P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">&nbsp;</FONT></FONT>     <BR><FONT SIZE=-1><FONT FACE="Arial,Helvetica">El n&uacute;mero esperado de fenotipos para cada enzima fue calculado asumiendo el equilibrio de Hardy Weimberg (<A HREF="#7">7</A>) y el estudio estad&iacute;stico de significancia se realiz&oacute; mediante la prueba de Chi cuadrado, para ello se utiliz&oacute; el programa ALFA del paquete GENIOC del Instituto Oswaldo Cruz de R&iacute;o de Janeiro, Brasil. La capacidad de un marcador gen&eacute;tico para distinguir entre individuos se relaciona con la frecuencia de sus fenotipos en una poblaci&oacute;n dada. El valor m&eacute;dico legal o forense de un marcador gen&eacute;tico se da por su capacidad de eliminar o discriminar entre individuos de una poblaci&oacute;n y para ello se debe calcular el poder de discriminaci&oacute;n (<A HREF="#8">8</A>).</FONT></FONT><FONT SIZE=-1></FONT>      <P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">Siendo Q la probabilidad de encontrar un pareo entre dos individuos seleccionados al azar usando un sistema gen&eacute;tico dado, conociendo las frecuencias p de los k fenotipos de este sistema tenemos que</FONT> <FONT FACE="Arial,Helvetica">(<A HREF="#8">8</A>):</FONT></FONT>     <BR><FONT SIZE=-1>&nbsp;</FONT><FONT SIZE=-1></FONT>     <CENTER><IMG SRC="/img/fbpe/rccm/v20n1-2/0642i1.GIF" HEIGHT=41 WIDTH=258></CENTER> <FONT SIZE=-1>&nbsp;</FONT><FONT SIZE=-1></FONT>

<BR><FONT SIZE=-1>&nbsp;</FONT><FONT SIZE=-1></FONT>      <P><B><FONT FACE="Arial,Helvetica"><FONT SIZE=-1>Resultados</FONT></FONT></B><FONT SIZE=-1></FONT>      <P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">Los valores de las frecuencias al&eacute;licas de los 10 sistemas enzim&aacute;ticos encontrados en la poblaci&oacute;n costarricense, y los valores de bondad de ajuste a la prueba de Chi cuadrado se presentan en los <A HREF="#CUADRO 1">cuadros 1</A> y <A HREF="#CUADRO 2">2</A>.</FONT></FONT><FONT SIZE=-1></FONT>      <P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">Los resultados del c&aacute;lculo de la probabilidad de discriminaci&oacute;n para cada uno de los sistemas estudiados se presentan en <A HREF="#CUADRO 3">cuadros 3</A> y <A HREF="#CUADRO 4">4</A>.</FONT></FONT>     <BR><FONT SIZE=-1>&nbsp;</FONT><FONT SIZE=-1></FONT>      <P><B><FONT FACE="Arial,Helvetica"><FONT SIZE=-1>Discusi&oacute;n</FONT></FONT></B><FONT SIZE=-1></FONT>      <P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">Las frecuencias fenot&iacute;picas encontradas est&aacute;n en equilibrio de Hardy Weimberg y los valores de Chi cuadrado no son significativos. Dado que la proporci&oacute;n de fenotipos heterocigotos encontrados guarda relaci&oacute;n con los esperados se puede afirmar que no se encontr&oacute; subestructura poblacional para estos marcadores en la muestra analizada. Los resultados obtenidos en la muestra de la poblaci&oacute;n general de Costa Rica son similares a los reportados en estudios poblacionales que incluyen grupos hispano-norteamericanos y cauc&aacute;sicos. Se puede citar que en grupos poblacionales de cauc&aacute;sicos e hispanos norteamericanos, la Adenosindeaminasa (ADA) presenta frecuencias de ADA*1=0.9483 y 0.9765 respectivamente (<A HREF="#8">8</A>,<A HREF="#9">9</A>) y en la muestra analizada encontramos una frecuencia de ADA*1= 0.966. En Glioxilasa, las frecuencias g&eacute;nicas reportadas en la poblaci&oacute;n cauc&aacute;sica europeos para Glo*1 y Glo*2 son iguales a las del presente estudio donde ambos genes se manifiestan en la poblaci&oacute;n con</FONT> <FONT FACE="Arial,Helvetica">frecuencias muy similares. Se muestra una franca diferencia de los valores reportados para Glo*1 en poblaciones japonesas, con frecuencias cercanas a 0.9 (<A HREF="#11">11</A>). Este fen&oacute;meno puede tambi&eacute;n observarse con la Anhidrasa carb&oacute;nico donde la frecuencia en cauc&aacute;sicos es de Ca*1= 0.9946 y la encontrada en el presente estudio es de Ca*1= 0.9946, valores que dan muy poco margen a la frecuencia del gen Ca*2 con valores que oscilan entre 0.0054 a 0.0213, y en poblaciones negras norteamericanas se ve una mayor frecuencia del gen Ca*2= 0.0972 (<A HREF="#9">9</A>,<A HREF="#10">10</A>).</FONT></FONT><FONT SIZE=-1></FONT>      <P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">Los polimorfismos gen&eacute;ticos son caracter&iacute;sticas gen&eacute;ticas que se encuentran en todas las poblaciones pero cuya frecuencia var&iacute;a entre ellas por lo cual han sido de gran utilidad en la caracterizaci&oacute;n de diferentes poblaciones y parece ser que la variaci&oacute;n dentro de los grupos poblacionales existentes actualmente es mayor que la existente entre grupos (<A HREF="#1">1</A>) tal y como lo demuestra la comparaci&oacute;n de los resultados obtenidos en el presente estudio con otros datos publicados.</FONT></FONT><FONT SIZE=-1></FONT>      ]]></body>
<body><![CDATA[<P><FONT SIZE=-1><FONT FACE="Arial,Helvetica">La individualidad bioqu&iacute;mica es notable cuando se consideran muchos de los polimortismos presentes en la sangre y se llega a detectar la heterogeneidad gen&eacute;tica existente entre los seres humanos.
